Effect of Sodium Tungstate on Nucleic Acid Extraction from Anaerobic Digestion Sludge —Efficacy of Fluorescence Measurement in the Ribonucleic Acid Quantification—

Nucleic acid extracted from environmental samples is an important analyte in instrumental analysis. With respect to the phenomenon which sodium tungstate promotes methane production anaerobic digestion, analyses of microbial community using RNA were conducted; however, we noticed that only concentration measured by absorbance measurement (NanoDrop) was remarkably high. Therefore, this study, DNA and digestion sludge with tungstate, selenite, or molybdate, these nucleic acids quantified compared measurement, fluorescence (Qubit), gel electrophoresis. Interestingly, it found sample containing NanoDrop 3 times higher than Qubit In addition, there no difference between Regarding other compounds, differences. Hence, results indicate system useful for quantification samples.
